That's a nice one. Probably a 1915 Babe Ruth, or a '85 Star Co Jordan myself.

It would definitely be something early vintage that even in pretty poor condition sells for Thou$and$, but of course with UNLIMITED Resources I would go as mint as currently possible. But what? With that kind of $$$ the imfamous T206 Honus Wagner comes to mind. However, if it were to be my ONLY card to buy I would get something else... Some later cards that are white whales are 1976 Topps Football Walter Payton, and the dreaded SSP 2012 Topps Archives Baseball 241 Bryce Harper RC come to mind, or the Michael Jordan RC but I still feel there is something bigger better I would get. As I said it would be something vintage. Maybe Walter Johnson's Rookie card.

Hmm...Probably not much different than it's going to be anyway, the next card from my want list that becomes available. My finds are few and far between lately, to the point that budget isn't much of a limiting factor. Of course my collection is very focused and I'm not much interested in expanding it's scope at this time.
If I suddenly came into unlimited resources (literally or in practical terms) my collecting focus would most likely shift towards coins and cars and maybe some actual memorabilia, as opposed to expanding my trading card want list.

Yeah, I like each of the last two answers. A vintage, nice Walter Johnson would be way up there. Or any vintage Babe Ruth.
But I'd might change my focus and buy some really cool piece if memorabilia. Not sure what that'd be.
Not to totally be a bummer, but I have trouble spending a ton of cash on one piece of cardboard. I recently had enough money gifted to me for the express purpose of buying baseball cards to buy one great card. It was a few hundred dollars. I couldn't do it. I couldn't even spend all the money on collection-related stuff. I spent about 80% of it on a variety of stuff and had a great time. Then I stopped. I want to enjoy the hobby without feeling guilty about it. So even with unlimited resources, I'd struggle to go nuts on a card. Maybe it's my poverty law background.
Whoa, I was, in fact, a total bummer. Sorry.
The Johnson rookie.

One card, unlimited resources. A Gem Mint 10, 1968 Topps #177 Jerry Koosman/Nolan Ryan rookie.

Someday my cards may double in value and then be worth half of what I paid for them.

I also thought of that as a possibility. Except I'm fine with an ungraded one that looks like a ten. Or a nine. Because I really wouldn't notice the difference.
